In Vitro Culture of Bone Marrow Cells (CFU-GM) from Drug-Induced Agranulocytosis

Abstract

|
A case with agranulocytosis following medication of anti-psychotic drugs was
presented. Bone marrow section revealed marked hypocellularity and myeloid cells not
visible. Bone marrow cells from this patient was cultured in vitro using semisolid agar
method with human placental conditioned medium as colony stimulating factor.
The results was as follows;
1) In control group, number of cluster was 119∼396/2×105 marrow cells
and that of colony was 20∼198/2×105 marrow cells. The cluster/colony
ratio was 4.8±1.9 (Mean±S.D., 1.7∼8.6).
2) In patient with Agranulocytosis, number of cluster was 108 and that of colony was
23. The cluster/colony ratio was 4.7. This result suggested that recovery from bone
marrow failure may be predicted before clinical findings are improved.
